    Now in the modern world Nonalcoholic fatty liver disease (NAFLD) is expected to be a major disorder in humans, insulin therapy is the main treatment meant for stabilizing the autoimmune disease and diabetes.

    The results on diabetes research expecting approximate of 592 million people will be becoming victims by the year 2050. Diabetes and its concern autoimmunity playing a major role for mitophagy, a metabolic disease with relevance to NAFLD epidemic. Autoimmune disease, drugs, and immunosenescence are related to chronic disease, which results in uncontrolled inflammatory cytokines such as tumor necrosis and interleukin.

    The major issue in global chronic disease is the autoimmune disease with defective adipose tissue, due to the release of inflammatory cytokines will effect lithe ver and adipocytokines relevant to toxic immune reactions that involve the pancreas, brain, heart, thyroid, kidneys and reproductive organs.     The state of insulin secretion is related to hyperglycemia, high serum low-density lipoprotein cholesterol concentrations and low serum high-density with relevance to cardiovascular disease. NAFLD may be connected to autoimmune disease and mitophagy associated with impairment in insulin secretion and cardiovascular disease. The research says that the diet, lifestyle, stress, sleep, and circadian disorders may cause to inactivate the anti-aging gene Sirtuin 1 (Sirt 1) with relevance to insulin therapy and induction of NAFLD associated with the pathogenetic loop and uncontrolled inflammation of cells and tissues. Exercise and insulin therapy may reduce glucolipotoxicity and NAFLD but with the aging process the pathogenetic loop that involves hyperglycemia, hypercholesterolemia and hyperinsulinemia may be associated with autoimmune disease, mitophagy and programmed cell death of various cells and tissues.

    The fatty liver acid metabolism related to mitochondrial dysfunction associated with severe liver inflammation and steatohepatitis that may induce NAFLD, in some cases it causes to a severe inflammation (NASH) and liver cirrhosis. Based on the study the Insulin therapy was used to improve liver function but with NAFLD, and also in case of high dose insulin therapy may be unsuccessful with liver inflammation. The diabetes of types-1,2 & 3 with liver disease medications are under study on Insulin therapy and oral anti-diabetic.

    The study says that uncontrolled diet may be critical to control the appetite for maintaining mitochondrial biogenesis and insulin therapy, also leads to severe diabetic conditions. Mainly the Indian spices have become important as a diabetes technology, spices such as curcumin and cinnamon associated with glucose control in diabetics. Also in some cases, the consumption of huge spices may interfere with insulin and oral medication therapy.

The major limitation with insulin therapy is to correlate the dose of insulin injected with plasma biomarkers that maintain mitochondrial biogenesis associated with the prevention of NAFLD. As the part of the study, antimicrobials use with insulin therapy should be carefully controlled to prevent the increased release of gram-negative bacteria lipopolysaccharides end products that may interfere with glucose homeostasis and induce NAFLD. The present study helps to determine the severity of diabetes with lipidomic analysis.

    Conclusion

    Insulin treatment has been evaluated in diabetes but the global NAFLD epidemic that is expected to reach between 20 and 30% of the worldwide communities will now be connected to diabetes pandemic and the pathogenetic loop. Insulin therapy with Indian spice consumption requires reassessment to avoid over-consumption of Indian spices that may inactivate insulin therapy and mitochondrial biogenesis. Food quality, appetite control, and core body temperature are critical to maintaining insulin therapy with unhealthy diets linked to NAFLD and diabetes. Insulin therapy may not reverse the nuclear-mitochondria defect that is relevant to global organ disease and various plasma biomarkers.

The researches in 1962 by Gowens demonstrated that lymphocytes proliferated in vivo after antigenic simulation, and later in 1963 and 1964, Kurt Hirschhorn Fritz Bach extended the experiments on specific antigen in vitro. These findings made the prediction from Burnet that antigen selected lymphocytes could undergo proliferative clonal expansion, a reality. Furthermore, extensive research studies were conducted by many researchers such as Neils Jerne, Jacques Miller, and Max Cooper in the 1960’s. By 1965, adaptive antigen-specific immunity was established to derive from small lymphocytes that originated in the thymus. The first reports of antigen-non-specific mitogenic activities produced by alloantigen-stimulated leukocytes were also released in 1965.
